چکیده

The pholyphenolic phytoalexin resveratrol (3,4 ́,5-trihydroxystilbene) is abundant in grape skins (with higher concentration in red as compared to white wine), peanuts, the roots of the weed Polygonum cuspidatum as well as in other fruits and vegetables (Cardile et al., 2007; Csaki et al., 2008). It is produced by plants in response to stress and has a wide range of pharmacological activities. Resveratrol possesses several pharmacological activities, such as anti-infl ammatory (Csaki et al., 2008; Bereswill et al., 2010), antiproliferative (Cardile et al., 2007; Gao et al., 2001, Glehr et al. 2013), pro-apoptotic (Byun et al., 2008; Glehr et al., 2013), antioxidant (Bhat et al., 2001), and cardioprotective (Lakota et al., 2009), with no obvious toxicity (Jang et al., 1997; Bhat et al., 2001; Baur and Sinclair, 2006). Rheumatoid arthritis (RA) is a chronic infl ammatory autoimmune disease characterized by synovial proliferation leading to cartilage and bone destruction (Lee et al., 2006). Contributing to the local production of cytokines, fi broblastlike synoviocytes (FLS) play a pivotal role in the pathogenesis of RA (Sipe et al., 1994; Burger et al., 2006). The influence of resveratrol on the synovial expression of matrix metalloproteinases and receptor activator of NF-kappaB ligand in rheumatoid arthritis fibroblast-like synoviocytes.

Medication of rheumatoid arthritis (RA) remains challenging and often controversial concerning side effects or long-term complications. We investigated the effect of resveratrol, a phytoalexin discussed for its chondro-protective and anti-inflammatory qualities, on the synovial expression of matrix-degrading enzymes like matrix metalloproteinases (MMPs) and bone-remodelling proteins in RA fibro...

Hypericin-photodynamic therapy inhibits proliferation and induces apoptosis in human rheumatoid arthritis fibroblast-like synoviocytes cell line MH7A

Objective(s): To elucidate the effects and potential mechanisms of hypericin-photodynamic therapy (HYP-PDT) for treating the human rheumatoid arthritis (RA) fibroblast-like synoviocyte (FLS) MH7A cell-line. Materials and Methods: MH7A cells were subjected to HYP-PDT intervention and apoptosis was evaluated via MTT, nuclear staining, and flowcytometry analyses. Intracellular reactive oxygen spec...

Celastrol Inhibits Lipopolysaccharide-Stimulated Rheumatoid Fibroblast-Like Synoviocyte Invasion through Suppression of TLR4/NF-κB-Mediated Matrix Metalloproteinase-9 Expression

Invasion of fibroblast-like synoviocytes (FLSs) is critical in the pathogenesis of rheumatoid arthritis (RA). The metalloproteinases (MMPs) and activator of Toll-like receptor 4 (TLR4)/nuclear factor-κB (NF-κB) pathway play a critical role in RA-FLS invasion induced by lipopolysaccharide (LPS). The present study aimed to explore the anti-invasive activity of celastrol on LPS-stimulated human RA...

Down-regulating peroxisome proliferator-activated receptor-gamma coactivator-1beta alleviates the proinflammatory effect of rheumatoid arthritis fibroblast-like synoviocytes through inhibiting extracellular signal-regulated kinase, p38 and nuclear factor-kappaB activation

INTRODUCTION Rheumatoid arthritis (RA) is a chronic inflammatory disease leading to joint destruction and disability. Peroxisome proliferator-activated receptor-gamma coactivator-1 beta (PGC-1β) is a transcriptional coactivator that plays important roles in regulating multiple aspects of energy metabolism and cytokine signaling pathways. PGC-1β overexpression leads to the attenuation of macroph...

Inhibition of microRNA-21 decreases the invasiveness of fibroblast-like synoviocytes in rheumatoid arthritis via TGFβ/Smads signaling pathway

Objective(s): MicroRNA-21 (miR21) is aberrantly elevated in rheumatoid arthritis (RA) patients, the significance of this microRNA in RA pathogenesis and treatment, however, has not been investigated. In this study, by using RA-derived fibroblast-like synoviocyte (FLS) cells as a model, we investigated the effect and corresponding mechanism of miR21 inhibition on FLSs invasion. Materials and Met...
